What is the standard tuning for bass?

The standard design for the electric bass guitar has four strings, tuned E, A, D and G, in fourths such that the open highest string, G, is an eleventh (an octave and a fourth) below middle C, making the tuning of all four strings the same as that of the double bass (E1–A1–D2–G2).

Does it matter if a guitar is left or right-handed?

Do you tune an electric guitar with the amp on or off?

As far as I know almost all tuner pedals mute the guitar signal when they’re activated. You push the foot-switch and it mutes your signal, no need to turn your amp on or off. However if you use a clip-on tuner you can just use a latching mute pedal while tuning your guitar.
